@brian, I see there is a service package to complete this task for me, is there anything missing from that or will it provide me with a fully functional port of my Ning site? Should/can I call someone to discuss it before purchase?

If you can create your Ning archive and upload it to your Jamroom server, alongside the Jamroom install (as per the Ning2JR import docs), we can then take over and do the rest of the import for you. All compatible data in your archive will be imported (assuming it is error free) and you'll be left with a ready to go Jamroom version of your Ning2 site based on the Ningja skin.

url mapper strictness

Actually, what you are trying works for me even without the URL Mapper module.

/firstname-lastname
/firstname_lastname
/firstname_lastname/

all take me to /firstname-lastname so this must be inherent within JR

Profile Domain Module - Procedure not Recommended?

I normally just point to the server IP, enable in JR then wait.
Sometimes after setting up it shows the default server page ("You have reached etc etc") instead of the correct JR profile. When that happens, I restart apache and it fixes things from then on.

Ning To Jamroom

Now back from a long and computer free weekend away and am faced with literally 100s of Jamroom notification emails.
It'll take me ages to go though them all, and hopefully a lot of the issues might have been solved, so can I ask that if anyone has issues, questions etc. that do want my attention now, just post here with a link to the thread, support ticket whatever, and I'll do my best to catch up.

Another option might be just to bring over your members and give them a clean slate to work with.
Members are your most valuable asset and arguably everything they've posted over the years is disposable.
I've been running a Jamroom site for over 11 years now and I've often thought about having a clear out of all the old stuff that is never read, watched or listened to any more, if only to get a load of disk space back!!

There is a Invite module coming soon. That will message followers and/or a list of email addresses inviting them to follow you, like a song/video/blog or generally get them to a specific page.
